Abstract
Ciljevi istraživanja: Razviti metodu za kvalitativno i kvantitativno određivanje metadona i EDDP-a u biološkim uzorcima korištenjem HPLC tehnike, koja se može primijeniti u stvarnoj praksi određivanja nepoznate koncentracije metadona u pacijenata na supstitucijskoj terapiji metadonom. Materijali i metode: Metodom HPLC su analizirane standardne otopine metadona i EDDP-a, pripremljene razrjeđivanjem standarda metadona i EDDP-a (Lipomed) te otopine Heptanona u metanolu. Nakon što su metadon i EDDP detektirani u standardnim otopinama te nakon ekstrakcije oba analita iz seruma korištenjem tehnike LLE (engl. liquid-liquid extraction), ekstrahirani analiti (smjesa metadona i EDDP-a) su u koncentraciji od 1 mg/L i volumenu od 10 µL injektirani u aparat. Optimalna valna duljina je bila 220 nm, kao pokretna faza je korištena smjesa metanola, acetonitrila i vode (v/v/v=9:11:80), pH= 6.0, temperatura sustava je namještena na 30°C, a vrijeme trajanja analize iznosilo je 8 minuta. Protok je bio namješten na 0.6 mL/min do četvrte minute pa na 0.4 mL/min od četvrte do osme minute. Rezultati: Injektiranjem standardne otopine metadona u prethodno navedenim uvjetima, kao i standardne otopine EDDP-a, oba su spoja na instrumentu detektirana oko šeste minute. Ipak, analiti ekstrahirani iz seruma (smjesa metadona i EDDP-a) su u istim uvjetima na kromatogramu detektirani kao jedan pik, umjesto dva odvojena pika koja su trebala odgovarati dvjema različitim tvarima. Zaključci: Analizom u prethodno obrazloženim uvjetima metadon i EDDP su kvalitativno dokazani, tj. dokazano je njihovo prisustvo u serumu. Ipak, navedenom metodom nije ih moguće dokazati kvantitativno, što znači da se ona ne može primijeniti u stvarnoj praksi određivanja nepoznate koncentracije lijeka i njegovih metabolita u biološkom uzorku.
Abstract (english)
Objectives: The aim of this study was to develop an analytical method for the HPLC determination of methadone and EDDP in biological fluids, which could be readily applied to the monitoring of methadone levels in biological fluids of patients undergoing methadone maintenance therapy. Materials and methods: Methadone and EDDP standard solutions were preapared after diluting the Heptanon solution, and both methadone and EDDP reference standards from Lipomed in methanol and then injected in the HPLC. After the methadone and EDDP standard solutions were detected on the instrument and both of the analites were extracted from serum by LLE, 10 µL of the extracted substances (the mixture of methadone and EDDP) was injected in the concetration 1 mg/L. The chromatografic procedure was carried out by setting a detection wavelength at 220 nm, the working temperature at 30°C and using mixture of methanol, acetonitrile and water (v/v/v=9:11:80), pH= 6.0, as mobile phase. The analysis was performed for 8 minutes total. The flow rate of the mobile phase was 0.6 mL/min during the first four minutes and then switched to 0.4 mL/min from the fourth minute to the end of the analysis. Results: Both of methadone and EDDP standard solutions were eluted around sixth minute. Still, the mixture of methadone and EDDP extracted from the serum was detected as only one peak on the chromatogram, instead of two separate peaks which should have correspond to these two different substances. Conclusion: Using the HPLC method described herein, the presence of both methadone and EDDP in the serum could be detected. However, their concentration could not be quantified, therefore this method could not be used for the determination of a drug and its metabolites in biological fluids.
