Abstract
Zbog brojnosti i kemijske raznolikosti molekula antioksidansa, različitih mehanizama njihova djelovanja, kao i ciljanih molekula na koje se želi ispitati njihov utjecaj, do danas je razvijen veliki broj antioksidacijskih metoda. Kako se niti jedna od postojećih metoda nije pokazala prikladnom da u potpunosti reflektira antioksidacijski kapacitet i svojstva uzorka, uobičajeno je da se u istraživanjima koristi nekoliko metoda koje se temelje na različitim reakcijskim mehanizmima. Ipak, veliki broj metoda i njihove modifikacije često dovode do nepravilne interpretacije rezultata i njihove otežane rezultata pa se javlja potreba za standardiziranom metodom koja bi omogućila jednoznačno mjerenje antioksidacijskog kapaciteta uzoraka. ORAC (engl. Oxygen Radical Absorbance Capacity) metoda je općeprihvaćena standardizirana antioksidacijska metoda. Prema mehanizmu reakcije koji se odvija kod ORAC metode ona spada u antioksidacijske metode koje se temelje na prijenosu atoma vodika. Rezultati, odnosno dobivene ORAC vrijednosti, uobičajeno se izražavaju preko Troloks ekvivalenta, u jedinicama µ M TE/g ili µ M TE/L uzorka, što znatno olakšava usporedbu rezultata različitih istraživanja.
Abstract (english)
Due to the abundance and chemical diversity of antioxidant molecules, their different mechanisms of action, as well as the final target molecules, a large number of antioxidant methods have been developed. As one antioxidant method can not describe the antioxidant properties and total antioxidant capacity of the sample, multiple-method approach, using methods based on different reaction mechanisms, is usually performed. However, a great number of methods and their modifications often lead to an incorrect interpretation of the results and caused problems in their comparison. Therefore, there is a need for a standardized method that would allow an unambiguous determination of antioxidant capacity of the samples. ORAC (Oxygen Radical Absorbance Capacity) assay is widely accepted and standardized antioxidant method. The mechanism of the reaction that takes place in the ORAC method is hydrogen atom transfer. The obtained results, or so called ORAC values, are usually expressed as Trolox equivalents, in µ M TE/g or µ M TE/L, what greatly allows the comparison of results from different studies.
